Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.411C>G (p.His137Gln), citing Ambry Variant Classification Scheme 2023: The p.H137Q variant (also known as c.411C>G), located in coding exon 4 of the TSC2 gene, results from a C to G substitution at nucleotide position 411. The histidine at codon 137 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
