Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_002087.4(GRN):c.128G>A (p.Arg43His), citing Ambry Variant Classification Scheme 2023: The p.R43H variant (also known as c.128G>A), located in coding exon 1 of the GRN gene, results from a G to A substitution at nucleotide position 128. The arginine at codon 43 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
