NM_002863.5(PYGL):c.607T>G (p.Phe203Val) was classified as Uncertain significance for Glycogen storage disease, type VI by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with valine at codon 203 of the PYGL protein (p.Phe203Val). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and valine. This variant has not been reported in the literature in individuals with PYGL-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt PYGL protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
