Uncertain significance for T-cell immunodeficiency, congenital alopecia, and nail dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001369369.1(FOXN1):c.1628G>A (p.Gly543Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 543 of the FOXN1 protein (p.Gly543Glu). This variant is present in population databases (rs752558563, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with FOXN1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1517581). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect FOXN1 function (PMID: 37419334). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
