Uncertain significance for Developmental and epileptic encephalopathy, 34 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020708.5(SLC12A5):c.2678T>C (p.Met893Thr), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with SLC12A5-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant is present in population databases (rs200461513, ExAC 0.009%). This sequence change replaces methionine with threonine at codon 893 of the SLC12A5 protein (p.Met893Thr). The methionine residue is highly conserved and there is a moderate physicochemical difference between methionine and threonine.
