NM_006118.4(HAX1):c.480G>C (p.Trp160Cys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W160C variant (also known as c.480G>C), located in coding exon 3 of the HAX1 gene, results from a G to C substitution at nucleotide position 480. The tryptophan at codon 160 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
