NM_014714.4(IFT140):c.1553C>T (p.Ser518Leu) was classified as Uncertain significance for Saldino-Mainzer syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces serine with leucine at codon 518 of the IFT140 protein (p.Ser518Leu). The serine residue is moderately conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs761365882, ExAC 0.002%). This variant has not been reported in the literature in individuals with IFT140-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
