Uncertain significance for Progressive sclerosing poliodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002693.3(POLG):c.280A>T (p.Met94Leu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the POLG gene (transcript NM_002693.3) at coding-DNA position 280, where A is replaced by T; at the protein level this means replaces methionine at residue 94 with leucine — a missense variant. Submitter rationale: This sequence change replaces methionine with leucine at codon 94 of the POLG protein (p.Met94Leu). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and leucine. This variant is present in population databases (rs778753625, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with POLG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
