NM_004984.4(KIF5A):c.2953G>T (p.Gly985Cys) was classified as Uncertain significance for Spastic paraplegia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt KIF5A protein function. This variant has not been reported in the literature in individuals with KIF5A-related conditions. This variant is present in population databases (rs371548640, ExAC 0.003%). This sequence change replaces glycine with cysteine at codon 985 of the KIF5A protein (p.Gly985Cys). The glycine residue is moderately conserved and there is a large physicochemical difference between glycine and cysteine.
