Uncertain significance for Primary familial hypertrophic cardiomyopathy; Dilated cardiomyopathy 1AA — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001103.4(ACTN2):c.1531C>G (p.Leu511Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with valine at codon 511 of the ACTN2 protein (p.Leu511Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACTN2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
