Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030957.4(ADAMTS10):c.1586G>A (p.Gly529Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 529 of the ADAMTS10 protein (p.Gly529Glu). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This missense change has been observed in individual(s) with Weill-Marchesani syndrome (PMID: 25053872). It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C65"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
