The leucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. ClinVar contains an entry for this variant (Variation ID: 1514944). This variant has not been reported in the literature in individuals affected with HMCN1-related conditions. This variant is present in population databases (rs772584982, gnomAD 0.04%), and has an allele count higher than expected for a pathogenic variant. This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 2867 of the HMCN1 protein (p.Val2867Leu). This variant also falls at the last nucleotide of exon 55, which is part of the consensus splice site for this exon. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
