Pathogenic for Epilepsy, childhood absence 4; Epilepsy, idiopathic generalized, susceptibility to, 13; Idiopathic generalized epilepsy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127644.2(GABRA1):c.755A>G (p.Tyr252Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 252 of the GABRA1 protein (p.Tyr252Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of GABRA1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1514692). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GABRA1 protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic.
