This sequence change replaces asparagine with serine at codon 527 of the CC2D2A protein (p.Asn527Ser). The asparagine residue is moderately conserved and there is a small physicochemical difference between asparagine and serine. This variant has not been reported in the literature in individuals affected with CC2D2A-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CC2D2A protein function.
