Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018474.6(KIZ):c.131A>G (p.Tyr44Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with cysteine at codon 44 of the KIZ protein (p.Tyr44Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with KIZ-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
