Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2255T>C (p.Leu752Pro), citing Ambry Variant Classification Scheme 2023: The p.L752P variant (also known as c.2255T>C), located in coding exon 14 of the ATM gene, results from a T to C substitution at nucleotide position 2255. The leucine at codon 752 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
