Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001235.5(SERPINH1):c.455A>G (p.Gln152Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with arginine at codon 152 of the SERPINH1 protein (p.Gln152Arg). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and arginine. This variant is present in population databases (rs768946201, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with SERPINH1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SERPINH1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
