Uncertain significance for Joubert syndrome 20; Meckel syndrome, type 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001077418.3(TMEM231):c.557G>T (p.Cys186Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with phenylalanine at codon 239 of the TMEM231 protein (p.Cys239Phe). The cysteine residue is weakly conserved and there is a large physicochemical difference between cysteine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TMEM231-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0").
