Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.2674C>T (p.Arg892Cys), citing Ambry Variant Classification Scheme 2023. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 2674, where C is replaced by T; at the protein level this means replaces arginine at residue 892 with cysteine — a missense variant. Submitter rationale: The p.R892C variant (also known as c.2674C>T), located in coding exon 19 of the DSP gene, results from a C to T substitution at nucleotide position 2674. The arginine at codon 892 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant has been detected in an exome sequencing cohort; however, details were limited (Yamaguchi-Kabata Y et al. J Hum Genet, 2018 Feb;63:213-230). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
