Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001298.3(CNGA3):c.2074A>C (p.Lys692Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine with glutamine at codon 692 of the CNGA3 protein (p.Lys692Gln). The lysine residue is moderately conserved and there is a small physicochemical difference between lysine and glutamine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CNGA3-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CNGA3 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
