Uncertain significance for Hereditary spastic paraplegia 74; Multiple mitochondrial dysfunctions syndrome 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001010867.4(IBA57):c.89G>A (p.Arg30His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 30 of the IBA57 protein (p.Arg30His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with IBA57-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The histidine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
