Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030631.4(SLC25A21):c.658A>G (p.Ile220Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 220 of the SLC25A21 protein (p.Ile220Val). This variant is present in population databases (rs765093427, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with SLC25A21-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
