This variant disrupts the extracellular domain of the CDH1 protein, which is important for cell-cell adhesion, correct folding, and dimerization (PMID: 8598933, 10439038, 10830618, 19268661, 22850631). While functional studies have not been performed to directly test the effect of this variant on CDH1 protein function, this suggests that disruption of this region of the protein is causative of disease. This variant has not been reported in the literature in individuals with CDH1-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 4-9 of the CDH1 gene.
