This variant lies in the NPR2 gene (transcript NM_003995.4) at coding-DNA position 1324, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 442 with asparagine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals affected with NPR2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with asparagine at codon 442 of the NPR2 protein (p.Asp442Asn). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and asparagine.
