Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001035.3(RYR2):c.3423+2T>C, citing Ambry Variant Classification Scheme 2023: The c.3423+2T>C intronic variant results from a T to C substitution two nucleotides after coding exon 28 in the RYR2 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. Variants that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this variant remains unclear.
