Uncertain significance for Dilated cardiomyopathy 1O — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020297.4(ABCC9):c.364T>G (p.Tyr122Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with aspartic acid at codon 122 of the ABCC9 protein (p.Tyr122Asp). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and aspartic acid. This variant has not been reported in the literature in individuals affected with ABCC9-related conditions. This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
