Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014862.4(ARNT2):c.1228T>A (p.Trp410Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 410 of the ARNT2 protein (p.Trp410Arg). This variant is present in population databases (rs150964641, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with ARNT2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1509647). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect ARNT2 function (PMID: 24465693). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
