Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.6814T>G (p.Phe2272Val), citing Ambry Variant Classification Scheme 2023: The p.F2272V variant (also known as c.6814T>G), located in coding exon 54 of the FBN2 gene, results from a T to G substitution at nucleotide position 6814. The phenylalanine at codon 2272 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
