This variant lies in the TRPM4 gene (transcript NM_017636.4) at coding-DNA position 416, where G is replaced by A; at the protein level this means replaces arginine at residue 139 with histidine — a missense variant. Submitter rationale: The p.R139H variant (also known as c.416G>A), located in coding exon 4 of the TRPM4 gene, results from a G to A substitution at nucleotide position 416. The arginine at codon 139 is replaced by histidine, an amino acid with highly similar properties. This variant was detected in an arrhythmia genetic testing cohort; however, clinical details were limited (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
