Uncertain significance for PTEN hamartoma tumor syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000314.8(PTEN):c.377C>G (p.Ala126Gly), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this variant affects PTEN protein function (PMID: 29987362, 26504226, 21828076). This variant has not been reported in the literature in individuals with PTEN-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with glycine at codon 126 of the PTEN protein (p.Ala126Gly). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and glycine.
