Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.3161A>T (p.Asp1054Val), citing Ambry Variant Classification Scheme 2023: The p.D1072V variant (also known as c.3215A>T), located in coding exon 14 of the MET gene, results from an A to T substitution at nucleotide position 3215. The aspartic acid at codon 1072 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
