Uncertain significance for Atypical hemolytic-uremic syndrome with MCP/CD46 anomaly — the classification assigned by 3billion to NM_172351.3(CD46):c.554A>G (p.Asp185Gly), citing ACMG Guidelines, 2015. This variant lies in the CD46 gene (transcript NM_172351.3) at coding-DNA position 554, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 185 with glycine — a missense variant. Submitter rationale: The variant is observed at an extremely low frequency in the gnomAD v4.1.0 dataset (total allele frequency: <0.001%). Predicted Consequence/Location: Missense variant. The majority of the known disease-causing variants of this gene are variants expected to result in premature termination of the protein. In silico tool predictions suggest damaging effect of the variant on gene or gene product [3Cnet: 0.95 (>=0.6, sensitivity 0.72 and precision 0.9)]. A different missense change at the same codon (p.Asp185Asn) has been reported to be associated with CD46-related disorder (PMID: 23431077). However the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
