Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001388492.1(HTT):c.9306G>C (p.Gln3102His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with histidine at codon 3104 of the HTT protein (p.Gln3104His). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and histidine. This variant is present in population databases (rs776079306, ExAC 0.006%). This variant has not been reported in the literature in individuals with HTT-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
