Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001244008.2(KIF1A):c.1726G>A (p.Val576Ile), citing Ambry Variant Classification Scheme 2023: The c.1699G>A (p.V567I) alteration is located in exon 19 (coding exon 18) of the KIF1A gene. This alteration results from a G to A substitution at nucleotide position 1699, causing the valine (V) at amino acid position 567 to be replaced by an isoleucine (I). Based on data from gnomAD, the A allele has an overall frequency of 0.003% (1/31386) total alleles studied. The highest observed frequency was 0.007% (1/15414) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
