Uncertain significance for Mitochondrial DNA depletion syndrome, encephalomyopathic form with methylmalonic aciduria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003850.3(SUCLA2):c.1112T>A (p.Leu371Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with glutamine at codon 371 of the SUCLA2 protein (p.Leu371Gln). The leucine residue is moderately conserved and there is a moderate physicochemical difference between leucine and glutamine. This variant is present in population databases (rs777781997, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with SUCLA2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
