Uncertain significance for Dyskeratosis congenita, autosomal dominant 2; Idiopathic Pulmonary Fibrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198253.3(TERT):c.2413C>T (p.Leu805Phe), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with phenylalanine at codon 805 of the TERT protein (p.Leu805Phe). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant has not been reported in the literature in individuals with TERT-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TERT protein function.
