Uncertain significance for Peroxisome biogenesis disorder 3A (Zellweger) — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000286.3(PEX12):c.191C>T (p.Thr64Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with isoleucine at codon 64 of the PEX12 protein (p.Thr64Ile). The threonine residue is moderately conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is present in population databases (rs766393993, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with PEX12-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
