NM_213599.3(ANO5):c.581A>G (p.His194Arg) was classified as Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2L; Gnathodiaphyseal dysplasia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ANO5 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with ANO5-related conditions. This sequence change replaces histidine with arginine at codon 194 of the ANO5 protein (p.His194Arg). The histidine residue is weakly conserved and there is a small physicochemical difference between histidine and arginine. This variant is present in population databases (rs146418790, ExAC 0.02%).
