Pathogenic for Epilepsy, idiopathic generalized, susceptibility to, 13; Idiopathic generalized epilepsy; Epilepsy, childhood absence 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127644.2(GABRA1):c.275T>C (p.Phe92Ser), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GABRA1 protein function. ClinVar contains an entry for this variant (Variation ID: 1506120). This missense change has been observed in individual(s) with clinical features of GABRA1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 92 of the GABRA1 protein (p.Phe92Ser).
