NM_153033.5(KCTD7):c.861_863del (p.Trp289del) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: KCTD7 c.861_863delATG (p.Trp288del) results in an in-frame deletion that is predicted to remove one amino acid from the encoded protein and the deletion of the last amino acid residue (tryptophan) of KCTD7. The variant was absent in 248190 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.861_863delATG has been reported in the literature in two affected siblings with progressive myoclonus epilepsies (Kousi_2012, Metz_2018). These reports do not provide unequivocal conclusions about association of the variant with Neuronal Ceroid-Lipofuscinosis (Batten Disease). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 22693283, 30295347, 34866617). ClinVar contains an entry for this variant (Variation ID: 1505073). Based on the evidence outlined above, the variant was classified as uncertain significance.
