Uncertain significance for Emery-Dreifuss muscular dystrophy 5, autosomal dominant — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182914.3(SYNE2):c.2362G>A (p.Glu788Lys), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1504696). This variant has not been reported in the literature in individuals affected with SYNE2-related conditions. This variant is present in population databases (rs767773345, gnomAD 0.003%). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 788 of the SYNE2 protein (p.Glu788Lys). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
