Uncertain significance for Congenital disorder of deglycosylation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018297.4(NGLY1):c.968T>C (p.Val323Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with alanine at codon 323 of the NGLY1 protein (p.Val323Ala). The valine residue is weakly conserved and there is a small physicochemical difference between valine and alanine. This variant is present in population databases (rs200601419, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with NGLY1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
