Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014822.4(SEC24D):c.95C>T (p.Pro32Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with leucine at codon 32 of the SEC24D protein (p.Pro32Leu). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and leucine. This variant is present in population databases (rs753308105, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with SEC24D-related conditions. ClinVar contains an entry for this variant (Variation ID: 1503305). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
