Submitter rationale: This variant has not been reported in the literature in individuals with CPLANE1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with lysine at codon 226 of the CPLANE1 protein (p.Arg226Lys). The arginine residue is weakly conserved and there is a small physicochemical difference between arginine and lysine. This variant also falls at the last nucleotide of exon 6, which is part of the consensus splice site for this exon.
