NM_004100.5(EYA4):c.961A>C (p.Asn321His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N321H variant (also known as c.961A>C), located in coding exon 10 of the EYA4 gene, results from an A to C substitution at nucleotide position 961. The asparagine at codon 321 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
