NM_000138.5(FBN1):c.7231G>A (p.Asp2411Asn) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015. This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 7231, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 2411 with asparagine — a missense variant. Submitter rationale: This missense variant replaces aspartic acid with asparagine at codon 2411 of the FBN1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been observed in an individual affected with aortic disease or Marfan syndrome (PMID: 27611364). This study has reported that this variant did not segregate with disease in the proband's family but details were not described. This variant has been identified in 4/251172 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
