Uncertain significance for Ehlers-Danlos syndrome, dermatosparaxis type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014244.5(ADAMTS2):c.2713C>T (p.Arg905Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ADAMTS2 gene (transcript NM_014244.5) at coding-DNA position 2713, where C is replaced by T; at the protein level this means replaces arginine at residue 905 with cysteine — a missense variant. Submitter rationale: This sequence change replaces arginine with cysteine at codon 905 of the ADAMTS2 protein (p.Arg905Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs760156480, ExAC 0.009%). This variant has not been reported in the literature in individuals with ADAMTS2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
