This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 1001, where G is replaced by T; at the protein level this means replaces glycine at residue 334 with valine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 334 of the IFIH1 protein (p.Gly334Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with IFIH1-related conditions. This missense change has been observed in at least one individual who was not affected with IFIH1-related conditions (Invitae).
