This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 1927, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 643 with lysine — a missense variant. Submitter rationale: Variant summary: ALMS1 c.1924G>A (p.Glu642Lys) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 248804 control chromosomes. c.1924G>A has been reported in the literature in at least one individual affected with Alstrom Syndrome. These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
