Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 251334 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.959A>G has been reported in the literature in the compound heterozygous state in at least one individual affected with CAD deficiency (DelCano-Ochoa_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Early Infantile Epileptic Encephalopathy, 50. At least one publication reports experimental evidence evaluating an impact on protein function showing that this variant showed a partial rescue in human CAD knockout cells (DelCano-Ochoa_2020).
